We are Isha &amp; Maia, the marine microbiologists on board the METEOR!<\/p>\n\n\n\n<p>Both of us are international students who left our home countries to pursue careers in science, specifically in microbial ecology, at the Carl von Ossietzky University of Oldenburg. We never dreamt that we would be conducting fieldwork for our PhDs in the middle of the Atlantic Ocean! Now that we\u2019re here, we\u2019d like to give you a glimpse into what we are doing on board and why.<\/p>\n\n\n\n<p>Our day starts with preparing the lab for all the incoming seawater samples, which come from the topmost layer or \u201cskin\u201d of the ocean, down to a depth of 600 m. We mount filters on six different filtration devices and lay out the tubes where these filters will eventually be stored. \u00a0During this preparation, our amazing colleagues are busy collecting the seawater for us. The sea surface microlayer (a widespread Surface-Film, which influences exchange processes between Ocean and Atmosphere) and the underlying water (one meter below the ocean surface) are collected by the Autonomous Surface Vehicle HALOBATES. This necessitates a lot of filters, which hold the answers to the questions of:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Which microbes live in these depths?<\/li>\n\n\n\n<li>What are they doing?<\/li>\n\n\n\n<li>How are they doing it?<\/li>\n<\/ul>\n\n\n\n<p>All of these questions will be answered throughout the following year using marker gene sequencing (16S rRNA analysis), sequencing of entire genomes of microbial communities (metagenomics), studying the proteins of these communities (metaproteomics), microscopy, and flow cytometry.<\/p>\n\n\n\n<figure class=\"wp-block-image size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"982\" height=\"736\" src=\"https:\/\/icbm-auf-see.uni-oldenburg.de\/wp-content\/uploads\/2025\/07\/Picture3-1.jpg\" alt=\"\" class=\"wp-image-3502\" srcset=\"https:\/\/icbm-auf-see.uni-oldenburg.de\/wp-content\/uploads\/2025\/07\/Picture3-1.jpg 982w, https:\/\/icbm-auf-see.uni-oldenburg.de\/wp-content\/uploads\/2025\/07\/Picture3-1-768x576.jpg 768w, https:\/\/icbm-auf-see.uni-oldenburg.de\/wp-content\/uploads\/2025\/07\/Picture3-1-640x480.jpg 640w\" sizes=\"auto, (max-width: 982px) 100vw, 982px\" \/><figcaption class=\"wp-element-caption\"><em>The filtration pumps are loud :\/ Hence the ear protection. \/\/ Die Filterger\u00e4te sind laut :\/ deshalb der Geh\u00f6rschutz.<\/em><\/figcaption><\/figure>\n\n\n\n<p>However, this \u201cleft-over\u201d, filtered seawater does not go to waste! A more specific question we utilize this water for is:<\/p>\n\n\n\n<p>How does the B vitamin availability and concentration differ in depth profiles of the North Atlantic?<\/p>\n\n\n\n<p>To answer this question, we perform solid-phase extraction, a method in which the filtrates are passed through cartridges that trap the B vitamins. There are teams studying the atmosphere, teams studying the physical aspects of the ocean, and our team studying the biological aspects of it. This may sound like several isolated stories, but it is quite the opposite. Our data is complementary, and we all contribute to one story, and we are glad to be a part of it.<\/p>\n\n\n\n<p>Deutsch<\/p>\n\n\n\n<p>Hi! Wir sind Isha &amp; Maia, die Meeresmikrobiologen an Bord der METEOR!<\/p>\n\n\n\n<p>Wir sind beide internationale Studenten, die ihre Heimatl\u00e4nder verlassen haben, um an der Carl von Ossietzky Universit\u00e4t Oldenburg eine wissenschaftliche Karriere in der mikrobiellen \u00d6kologie zu verfolgen. Nie h\u00e4tten wir gedacht, dass wir die Feldforschung f\u00fcr unsere Doktorarbeiten mitten im Atlantik betreiben w\u00fcrden, nun sind wir aber hier! Im folgenden geben wir einen Einblick in das, was wir an Bord tun und warum.<\/p>\n\n\n\n<p>Unser Tag beginnt mit der Vorbereitung des Labors f\u00fcr all die im Laufe des Tages ankommenden Meerwasserproben, die aus der obersten Schicht oder \u201eHaut\u201d des Ozeans bis zu einer Tiefe von 600 m entnommen werden. Als Vorbereitung montieren wir Filter auf sechs verschiedene Filterger\u00e4te und legen die R\u00f6hrchen aus, in denen diese Filter sp\u00e4ter aufbewahrt werden. W\u00e4hrend dieser Vorbereitungen sind unsere gro\u00dfartigen(!) Kollegen damit besch\u00e4ftigt, das Meerwasser f\u00fcr uns zu sammeln. Das Surface Microlayer (weitverbreiteter Oberfl\u00e4chenfilm, wodurch Austausch-Prozesse zwischen Ozean und Atmosph\u00e4re verlaufen) und das darunter liegende Wasser (in bis zu einem Meter unter der Meeresoberfl\u00e4che) werden vom autonomen Oberfl\u00e4chenfahrzeug HALOBATES gesammelt. Die anderen Tiefen werden mit Niskin-Flaschen gesammelt, die an einer CTD angebracht sind.<\/p>\n\n\n\n<p>An einer einzigen Station filtern wir insgesamt 120 Liter Meerwasser. Das bedeutet jede Menge Filter (!), die Antworten auf folgende Fragen enthalten:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Welche Mikroben leben in diesen Tiefen?<\/li>\n\n\n\n<li>Was tun sie?<\/li>\n\n\n\n<li>Wie tun sie das?<\/li>\n<\/ul>\n\n\n\n<p>All diese Fragen werden im Laufe des folgenden Jahres mithilfe der Markergen-Sequenzierung (16S-rRNA-Analyse), der Sequenzierung ganzer Genome mikrobieller Gemeinschaften (Metagenomik), der Untersuchung der Proteine in diesen Gemeinschaften (Metaproteomik), der Mikroskopie und der Durchflusszytometrie beantwortet.<\/p>\n\n\n\n<p>Dennoch wird das \u201e\u00fcbrig gebliebene\u201d, gefilterte Meerwasser nicht verschwendet! Eine spezifischere Frage, f\u00fcr die wir dieses Wasser verwenden, lautet:<\/p>\n\n\n\n<p>Wie unterscheiden sich die Verf\u00fcgbarkeit und Konzentration von B-Vitaminen in den Tiefenprofilen des Nordatlantiks?<\/p>\n\n\n\n<p>Um diese Frage zu beantworten, f\u00fchren wir eine Festphasenextraktion durch ein Verfahren, bei dem die Filtrate durch Kartuschen geleitet werden, die die B-Vitamine auffangen. Die Kartuschen werden aufbewahrt und sp\u00e4ter in unseren Labors in Oldenburg mittels Fl\u00fcssigchromatografie-Massenspektrometrie analysiert, wodurch die B-Vitamine quantifiziert und identifiziert werden.<\/p>\n\n\n\n<p>Obwohl die Stationen k\u00f6rperlich und geistig anstrengend sind, finden wir dennoch Zeit, die gem\u00fctlichen Abende mit den Menschen an Bord, die atemberaubende Aussicht auf den Ozean und das n\u00e4chtliche Sternegucken zu genie\u00dfen.<\/p>\n\n\n\n<p>Diese Expedition ist in Bezug auf die verschiedenen Arbeitsgruppen mit unterschiedlichen Forschungsfragen definitiv sehr vielf\u00e4ltig. Es gibt Teams, welche die Atmosph\u00e4re untersuchen, Teams, die physikalische Aspekte des Ozeans untersuchen, und unser Team, das die biologischen Aspekte untersucht. Das mag nach mehreren isolierten Geschichten klingen, aber das Gegenteil ist der Fall.
